Scientists pair blood test and gene sequencing to detect cancer

Scientists at the Johns Hopkins Kimmel Cancer Center have combined the ability to detect cancer DNA in the blood with genome sequencing technology in a test that could be used to screen for cancers, monitor cancer patients for recurrence and find residual cancer left after surgery.

"This approach uses the power of genome sequencing to detect circulating tumor DNA in the blood, providing a sensitive method that can be used to detect and monitor cancers," says Victor Velculescu, M.D., Ph.D., professor of oncology and co-director of the Cancer Biology Program at Johns Hopkins.

A report describing the new approach appears in the Nov. 28 issue of Science Translational Medicine. To develop the test, the scientists took blood samples from late-stage colorectal and breast cancer patients and healthy individuals and looked for DNA that had been shed into the blood.

The investigators applied whole-genome sequencing technology to DNA found in blood samples, allowing them to compare sequences from cancer patients with those from healthy people. The scientists then looked for telltale signs of cancer in the DNA: dramatic rearrangements of the chromosomes or changes in chromosome number that occur only in cancer cells.

No signs of cancer-specific chromosome changes were found in the blood of healthy individuals, but the investigators found various cancer-specific alterations in the blood of all seven patients with colon cancer and three patients with breast cancer. Using specialized bioinformatic approaches, they were able to detect these alterations in a small fraction of the millions of DNA sequences contained in the blood sample.

"This is proof of the principle that genome sequencing to identify chromosomal alterations may be a helpful tool in detecting cancer DNA directly in the blood and, potentially, other body fluids," says Rebecca Leary, a postdoctoral fellow at Johns Hopkins. "But larger clinical trials will be needed to determine the best applications of this approach."

The investigators note that there may be less circulating DNA in early stage cancers, and, thus, these would be more challenging to detect without more extensive sequencing. As sequencing costs decrease, the investigators expect that detecting earlier-stage cancers may become more feasible.

Velculescu says that additional research will focus on determining how the new test could help doctors make decisions on treating patients. For example, the blood test could identify certain chromosomal changes that guide physicians to prescribe certain anti-cancer drugs or decide patient enrollment in clinical trials for drugs that target specific gene defects. Currently, physicians use cellular material biopsied from the original tumor to make these decisions, but tumor material can often be inaccessible or unavailable.

The Johns Hopkins study builds on the team's earlier work using genomic sequencing of DNA in the blood to find rearrangements of chromosomes. The previous work required samples of the original tumor and knowledge of DNA changes in that tumor to find those same changes in the blood. This new test has no need for original tumor samples and includes an analysis of changes in the copy number of chromosomes.

"It's an evolution of technologies we're developing for cancer diagnosis, and, by combining our knowledge, we can build better ways to detect disease," says Luis Diaz, M.D., an oncologist and director of the Swim Across America laboratory at Johns Hopkins.

Two winners in record Powerball jackpot

Winning tickets for the record Powerball jackpot worth more than $579 million were purchased in Arizona and Missouri.

Missouri Lottery official Susan Goedde confirmed to ABC News this morning that one of the winning tickets was purchased in the state, but they would not be announcing a town until later this morning. Powerball's website reports the other winning ticket was purchased in Arizona.

The winning numbers for the jackpot were 5, 23, 16, 22 and 29. The Powerball was 6.

Before the numbers were drawn on Wednesday, the jackpot swelled to $579.9 million, making the cash option $379.8 million.

An additional 8,924,123 players won smaller prizes, according to Powerball's website.

"There were 58 winners of $1 million and there were eight winners of $2 million. So a total of $74 million," said Chuck Strutt, Director of the Multi-State Lottery Association.

Hopeful players bought tickets at the rate of 131,000 every minute up until an hour before the deadline of 11 p.m. ET, according to lottery officials.

The jackpot had already rolled over 16 consecutive times without a winner. That fact, plus the doubling in price of a Powerball ticket, accounted for the unprecedented richness of the pot.

"Back in January, we moved Powerball from being a $1 game to $2," said Mary Neubauer, a spokeswoman at the game's headquarters in Iowa. "We thought at the time that this would mean bigger and faster-growing jackpots."

That proved true. The total, she said, began taking "huge jumps -- another $100 million since Saturday." It then jumped another $50 million.

The biggest Powerball pot on record until now -- $365 million -- was won in 2006 by eight Lincoln, Neb., co-workers. As the latest pot swelled, lottery officials said they began getting phone calls from all around the world.

"When it gets this big," said Neubauer, "we get inquiries from Canada and Europe from people wanting to know if they can buy a ticket. They ask if they can FedEx us the money."

The answer she has to give them, she said, is: "Sorry, no. You have to buy a ticket in a member state from a licensed retail location."

About 80 percent of players don't choose their own Powerball number, opting instead for a computer-generated one. Asked if there's anything a player can do to improve his or her odds of winning, Neubauer said there isn't -- apart from buying a ticket, of course.

Lottery officials put the odds of winning the $579 Powerball pot at one in 175 million, meaning you'd have been 25 times more likely to win an Academy Award.

Skip Garibaldi, a professor of mathematics at Emory University in Atlanta, provided additional perspective: You are three times more likely to die from a falling coconut, he said; seven times more likely to die from fireworks, "and way more likely to die from flesh-eating bacteria" (115 fatalities a year) than you are to win the Powerball lottery.

Segueing, then, from death to life, Garibaldi noted that even the best physicians, equipped with the most up-to-date equipment, can't predict the timing of a child's birth with much accuracy.

"But let's suppose," he said, "that your doctor managed to predict the day, the hour, the minute and the second your baby would be born."

The doctor's uncanny prediction would be "at least 100 times" more likely than your winning.

Even though he knows the odds all too well, Garibaldi said he usually plays the lottery.

When it gets this big, I'll buy a couple of tickets," he said. "It's kind of exciting. You get this feeling of anticipation. You get to think about the fantasy."

So, did he buy two tickets this time?

"I couldn't," he told ABC News. "I'm in California" -- one of eight states that doesn't offer Powerball.

Ex-'Melrose' star guilty of vehicular homicide

Amy Locane-Bovenizer is taken into custody after her conviction for vehicular homicide.

Sadly, this is not a fictional TV drama for Amy Locane-Bovenizer. It's her life.?The former "Melrose Place" star was found guilty on Tuesday of vehicular homicide and assault by auto resulting from a fatal June 2010 DUI crash in Montgomery Township, NJ.

Per New Jersey's Star Ledger, the 40-year-old Locane was acquitted of the more serious charge of aggravated manslaughter when her 2007 Chevy Tahoe struck a 2010 Mercury Milan as it was turning into a driveway. The collision killed a 60-year-old female passenger, Helene Seeman, and seriously injured her husband, Fred Seeman.

"Melrose Place" alum Amy Locane-Bovenizer charged with vehicular homicide

Court papers later showed that the actress admitted to police that she had drank several glasses of wine before getting behind the wheel. Not only that: Before she hit the Seemans, she had already rear-ended a car a few miles from the accident site and sped off before the other driver could call the cops.

Immediately following her conviction, a judge revoked Locane-Bovenizer's bail and remanded her into custody to await sentencing scheduled for March 1.?

The Council of PR Firms offers a paper called, ?Procurement Professionals Q&A: Advancing the Working Relationship with Public Relations Firms? which proposes the money side be handled as thus:

With regard to the financial negotiation, there should be a short term and longer term goal. Immediately, the client organization should communicate the reason for retaining the public relations firm and how the firm?s success will be evaluated. Longer term, the focus should be on relationship management based on the scope of work, benchmarks against goals, and appropriate compensation. Each party should make the other aware of what it is seeking to achieve in the relationship. The most successful and productive long term relationships are based on mutual respect and open dialogue.

It all sounds so good ? cue the birds chirping in the background ? in theory.

Unfortunately, the reality rarely plays out this way, at least in our experiences of interacting with Procurement. Instead, Procurement?s overriding objective if not singular mission is to secure the PR agency?s services at the lowest price point possible. While the Council?s paper stresses that PR services are not a commodity, Procurement hasn?t got the memo typically treating the PR agency negotiation like an express mail contract.

I do understand the quest by every company in the galaxy to gain value from their outside purchases.

Everyone wants a good deal.

I get it.

But I also think Procurement misses the big picture in the sense that no matter how tightly you scope the assignment and squeeze the PR agency, the intangible called ?care? is a huge wildcard. If an agency cares about the client, it actually goes above and beyond the statement of work (what a concept.).

Assuming Procurement professionals read the Council?s paper, I?m not convinced anything would change. The core issue is not Procurement?s lack of understanding the world of PR. The core issue is a rewards system that motivates Procurement to pummel vendors into submission to save another nickel an hour.

Transposable elements reveal a stem cell specific class of long noncoding RNAs

ScienceDaily (Nov. 26, 2012) ? Over a decade after sequencing the human genome, it has now become clear that the genome is not mostly 'junk' as previously thought. In fact, the ENCODE project consortium of dozens of labs and petabytes of data have determined that these 'noncoding' regions house everything from disease trait loci to important regulatory signals, all the way through to new types of RNA-based genes.

Yet over 70 years ago, it was first proclaimed that all this junk wasn't so junky. Barbara McClintock discovered the first utility of all of this junk DNA: jumping genes, also known as transposable elements. These genes serve only one purpose, which is to replicate themselves and reinsert randomly in the genome, or do they? Ironically, at the same time two other scientists (Roy Britten and Eric Davidson) proposed that jumping genes may be involved in regulating cell specificity. Indeed, in an exciting new study published in Genome Biology, John Rinn and David Kelley based at Harvard University and the Broad Institute in Boston, USA, provide genome-wide evidence that jumping genes may shape when a gene is turned on or off in stem cells.

"We set out to investigate how jumping genes have invaded the genome to potentially give rise to new genes in the 'junk regions'" says Rinn, the senior author of the study. "It has become very clear that there are thousands of long intergenic noncoding RNA genes (lincRNAs) that may herald a new paradigm for human health and disease." Yet how these genes have evolved from such a desert wasteland has remained a burning question. A new clue has emerged from the jumping genes that compose nearly 50% of the human genome.

"I like to think of it as 'on the origins of lincRNAs'" says Rinn. "It doesn't take more than a brief survey of McClintock, Britten and Davidson's work in the 50s and 60s to realize that transposable elements were a great first place to look. The human genome is in a constant battle with transposable elements with them randomly hopping into new locations, for good or for bad." Kelley adds that "In my Ph.D. work assembling genomes from sequence fragments, these repetitive hopping genes were a major nuisance, which got me thinking about what they were doing in the genome." The study published by Rinn and Kelley finds a striking affinity for a class of hopping genes known as endogenous retroviruses, or ERVs, to land in lincRNAs. The study finds that ERVs are not only enriched in lincRNAs, but also often sit at the start of the gene in an orientation to promote transcription. Perhaps more intriguingly, lincRNAs containing an ERV family known as HERVH correlated with expression in stem cells relative to dozens of other tested tissues and cells. According to Rinn, "This strongly suggests that ERV transposition in the genome may have given rise to stem cell-specific lincRNAs. The observation that HERVHs landed at the start of dozens of lincRNAs was almost chilling; that this appears to impart a stem cell-specific expression pattern was simply stunning!"

These results also raise the tantalizing question of why transposable elements, derived from viruses, regulate stem cell-specific expression in mammals. Rinn hypothesizes that "transposable elements may not be limited to giving rise to new lincRNA genes, but may also provide an engine for the evolution of RNA-encoding genes. I like to think of it as the 'genome getting dirty': in the same way that kids that play in the dirt develop better immune systems, the genome may be 'getting dirty' with transposable elements, and once in a while, this has an advantageous effect of producing a new lincRNA gene."

What is clear is that transposable elements may control the tissue-specific expression of lincRNAs, thereby affecting the evolution and function of lincRNAs with important regulatory roles. Following on from these results, it will be interesting to determine other ways hopping genes may have shaped lincRNA evolution. Kelley notes that "This study merely scratches the surface of the possible roles of transposable elements influencing lincRNA function."

Keystroke-logger checks your identity as you type

WHETHER an aggressive finger-jabber or a fluent touch-typist, the way you type says more about you than you might think. A biometric authentication system monitors the telltale timing gaps between the letters you type to continually verify your identity.

The traditional password is notoriously troublesome as a way of keeping your devices secure. Many people use simple, easy-to-guess passwords like qwerty or 123456, or reuse the same one across multiple services, putting only one line of defence in front of their entire digital life.

David Hibler of Christopher Newport University, Virginia, and colleagues designed software called URIEL, which uses the average time between keystrokes as a surprisingly accurate way of identifying an individual. In order for your carbon footprint not to get any bigger, you will need to use the dryer, along with the electricity, as little as possible. Not only is a dryer hard on the environment, no matter if it is gas or electric, but static electricity is increased by using it, especially in the winter. When you hang dry your clothing, the environment isn?t the only winner. You can save money by not having to replace your clothing as often, because they will last longer not being banged around in the dryer.

A good reusable mop is an investment you should make. Many people switch to Swiffer?s which are an alternative to traditional mops. This is because the use once and toss them pads make floor cleaning a much easier and less time consuming chore. The sad fact is, though, that the chemicals used in the cleanser the pads are soaked in is dangerous for you and your family and the pads themselves contribute to the overflowing of landfills. A reusable mop allows you the freedom to use environmentally friendly floor cleaner and helps you save lots of money over the long haul. When you spend your time cleaning, it will be more profitable if you do the job the right way. Before you choose any home improvement project, start with a thorough cleaning. One of the best ways to improve your health is to start cleaning your house by using cleaning methods that are green.

We are a group of international students studying abroad in Minnesota. Yesterday we decided to have a couple of beers and got busted. No one was over 21 and a couples were under 18. (AP) ? Bella Swan, James Bond and Abe Lincoln have combined to lift Hollywood to record Thanksgiving revenue at the box office.

Kristen Stewart's finale as Bella in "The Twilight Saga: Breaking Dawn ? Part 2" was No. 1 again with $64 million during the five-day holiday stretch that began Wednesday, according to studio estimates Sunday.

Daniel Craig's Bond adventure "Skyfall" came in at No. 2 with $51 million, while Daniel Day-Lewis and Steven Spielberg's Civil War saga "Lincoln" finished third with $34.1 million.

According to box-office tracker Hollywood.com, the three films paced Hollywood to an all-time Thanksgiving week best of about $290 million from Wednesday to Sunday.

That tops the previous record of $273 million over Thanksgiving in 2009, when "The Twilight Saga: New Moon" led the weekend.

This Thanksgiving also was a huge 25 percent jump from a year ago, when domestic revenues were a weak $232 million as some big holiday releases fizzled.

With a strong December lineup ahead, Hollywood has resumed its record revenue pace for the year after a brief box-office lull in late summer and early fall.

Domestic revenues for 2012 are at $9.75 billion, putting Hollywood potentially on track for its first $11 billion year, which would beat the 2009 record of $10.6 billion, said Hollywood.com analyst Paul Dergarabedian.

"We're barreling toward a record-breaking box-office year," Dergarabedian said. "It's built on the back of just a lot of really strong movies that have come out over the past few weekends. It bodes very well for the rest of the holidays."

The "Twilight" finale, "Skyfall" and "Lincoln" finished in the same top-three rankings for the second-straight weekend as new releases were unable to dislodge the holdovers.

Released by Lionsgate's Summit Entertainment banner, "Breaking Dawn ? Part 2," pulled in $43.1 million from Friday to Sunday, raising its domestic total to $227 million. The movie added $97.4 million overseas to bring its international total to $350.8 million and its worldwide take to $577.7 million.

Sony's "Skyfall" also topped $200 million domestically, ringing up $36 million for the three-day weekend to put its U.S. total at $221.7 million. With $41.3 million more overseas, "Skyfall" raised its international revenues to $568.4 million and its worldwide sales to $790.1 million.

"Lincoln," a DreamWorks film distributed by Disney, took in $25 million over the weekend to lift its domestic revenue to $62.2 million.

Leading the newcomers was Paramount and DreamWorks Animation's tale "Rise of the Guardians" at No. 4 with $24 million for the weekend and $32.6 million since opening Wednesday.

Based on William Joyce's "Guardians of Childhood" books, "Rise of the Guardians" gathers Santa Claus, the Easter Bunny, the Tooth Fairy and other mythical beings as a team of heroes battling an evil overlord.

Close behind at No. 5 was director Ang Lee's shipwreck saga "Life of Pi" at No. 5 with $22 million over the weekend. The 20th Century Fox release has taken in $30.2 million domestically since its Wednesday debut and added $17.5 million in four Asian markets.

"Life of Pi" was adapted from Yann Martel's best-selling novel about an Indian youth adrift on a lifeboat with a Bengal tiger. Many fans considered the introspective novel impossible to film, but Lee has charmed audiences and critics with an inspiring survival story told through dazzling 3-D images.

The weekend's other new wide release, a remake of the 1980s U.S.-invasion tale "Red Dawn," opened at No. 7 with $14.6 million, raising its total to $22 million since debuting Wednesday.

"Red Dawn" sat on the shelf for three years while studio backer MGM went through bankruptcy, with distributor FilmDistrict eventually picking it up for domestic release. The movie's cast includes Chris Hemsworth ("Thor") and Josh Hutcherson ("The Hunger Games") in a story of young guerrillas battling North Korean invaders.

In limited release, Fox Searchlight's "Hitchcock" opened solidly with about $300,000 in 17 theaters. The movie stars Anthony Hopkins as Alfred Hitchcock in a behind-the-scenes story of the making of "Psycho."

The weekend's overall strength came from a broad range of films that clicked with various audiences, from action and family fare to thoughtful drama.

"This is a marketplace that has something for everyone," said Chris Aronson, head of distribution for 20th Century Fox. "You have something deeper like 'Life of Pi,' yet you have a very successful sequel in 'Twilight' at the same time. Adult bio-drama, if you will, in 'Lincoln,' and you have Bond. That's the secret to a very successful and balanced marketplace."

Estimated ticket sales for Friday through Sunday at U.S. and Canadian theaters, according to Hollywood.com. Where available, latest international numbers are also included. Final domestic figures will be released Monday.
